Research Interests

Amyotrophic lateral sclerosis (ALS), also known as Lou Gehrig's disease, is a lethal progressive, neurological disorder affecting motor neurons. To find new therapies for ALS, Dr. Mougeot is developing in vitro cellular models for high-throughput screening (HTS) of small compounds, small interfering ribonucleic acid (siRNA) and monoclonal antibody libraries. Dr. Mougeot is using the mouse embryonic stem cell line HBG3 to produce GFP-expressing motor neurons. These motor neurons will be used to establish in vitro motor neuron / muscle cell cultures to conduct HTS assays and assays designed to better understand mechanism of cell death occurring early and late in the disease. Currently, early diagnosis of ALS provides the best hope of delaying disease progression with appropriate therapy and improving the quality of life in a timely manner. Dr. Mougeot is currently implementing validation studies following the determination of gene expression profiles inlymphocytes from patients with definite ALS compared to healthy controls. Potential biomarkers will subsequently be confirmed in the sera of ALS patients or studied for their relation with ALS pathogenesis using cell based assays.
